内容記述 Fungal contamination at cultural properties has been a worldwide problem. We have carried out our research on the measures for fungus contamination as one of the projects of conservation and restoration of cultural properties in Egypt, which is supported by the Ministry of Education, Culture, Sports, Science and Technology, Japan. In this study, we identified several fungus specimens isolated from Egyptian cultural properties and their surrounding space, and then characterized the growth of representative xerophilic fungi among them on the painted stone chips, supposing the prevention from fungi of mural painting in hypogeum of Idout Mastaba in Saqqara, Egypt. As a result of the identification of isolated 19 fungus specimens, xerophilic fungi were dominant, being 11 of Aspergillus restrictus and 6 of Eurotium amstelodami. Based on this finding, we examined the properties of the development of spores of these two fungal species by using laboratory model mural paintings. A stone chip was prepared and painted zonally with different pigments mixed with Arabic gum or animal glue as an adhesive. A small filter paper disk dipped with M40Y medium was inoculated with the fungal spore suspension, and the paper disk was placed onto the stone chip. After that, the stone was placed in a transparent plastic case in which each plastic petri dish contained a saturated solution of potassium nitrate to adjust a relative humidity of 95%, and thereafter the plastic case was incubated at 25 ℃ and at a relative humidity of 95%. The spore development and the subsequent growth were evaluated by measuring the diameter of colony formed on the stone chip. The results obtained were as follows. 1) Under tested conditions, in general, E. amstelodami grew better than A. restrictus. 2) As for the adhesive, in both species, Arabic gum supported better growth at 15% than at 30% as a concentration of solution at the painting, while in animal glue the situations varied according to the kind of pigment. 3) In the effect of the kind of pigment, A restrictus grew well particularly on brownish earth pigment with 15% Arabic gum and E. amstelodami grew well on blackish carbon with both adhesives and on yellowish earth pigment with animal glue at 30%. Conversely, the growth of A. restrictus was slightly suppressed by greenish Malachite with both adhesives, but we detected no definite trend for E. amstelodami. 4) In a separate experiment, we compared the fungal growth-inhibitory activity on different pigments, which were immixed in M40Y agar plates at concentration ratios used in the above painted chip experiment. As a result, against both fungi, blue and green pigments were found to be the most toxic, followed by yellow one, but red and black pigments were much less toxic. From the results obtained, we discussed the growth behaviors of xerophilic fungi on the stone chips painted with different pigments mixed with adhesives, and we infer conditions and situations on actual mural painting in possible fungal contamination.
